NAD+ Precursor Chews vs. Fish Oil for Senior Dog Vitality: What the Label and the Bloodwork Actually Show

A twelve-year-old Cavalier named Pepper came into my exam room because her owner had two bags on the counter and couldn’t decide which one to open first. One promised “cellular energy and longevity.” The other promised “vitality and joint comfort.” Pepper herself just wanted breakfast. I flipped both bags over before I said a word, because that’s where the actual answer lives — not on the front panel.
Here’s the checklist I actually use before I tell an owner which one, or both, or neither, makes sense for their dog:
- What’s the diagnosis, if any? Cognitive slowing and joint stiffness point in different directions.
- What’s the actual dose per pound, not per scoop? A “proprietary blend” tells you nothing.
- Is the NAD+ precursor named specifically — nicotinamide riboside (NR) or nicotinamide mononucleotide (NMN) — or just “NAD+ support”?
- Is the fish oil measured in EPA+DHA milligrams, not total fish oil milligrams? Those numbers are not the same thing.
- Has a controlled trial in dogs, not rodents or marketing copy, actually tested this combination?
- Is there a NASC seal or third-party certificate of analysis, or just a claim of one?

What NAD+ Precursors Are Actually Doing at the Cellular Level
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme every cell needs to run its energy metabolism, and levels measurably fall with age. Chews marketed for “cellular energy” or “longevity” are almost always delivering a precursor — NR or NMN — on the theory that more raw material means more NAD+ downstream. That’s a real, testable claim, and unlike a lot of longevity marketing, it’s actually been tested in dogs.
A randomized, controlled trial published in Scientific Reports gave a senolytic-and-NAD+-precursor combination to senior dogs with mild to moderate cognitive impairment and tracked owner-assessed cognitive decline scores over the study period. The dogs on the full dose showed the largest improvement relative to placebo. That’s a meaningfully different evidence tier than most supplement claims I read — it’s a real RCT in the actual species, with an actual comparator group, not an extrapolation from a mouse study or a testimonial page.
What it doesn’t tell you is whether NAD+ precursors do anything for joint comfort, coat, or general “vitality” in a dog that isn’t cognitively declining. The trial measured cognition. If your dog’s issue is a stiff walk to the mailbox rather than a foggy stare at the wall, this data point doesn’t transfer.
Where Fish Oil Still Does the Heavy Lifting
Marine omega-3s — EPA and DHA specifically, not “fish oil” as a vague category — have the deepest and widest evidence base of almost anything on a supplement shelf for dogs, and the dosing has actually been standardized across conditions. A review in PubMed pooling companion-animal omega-3 trials found effective ranges of roughly 48–100 mg/kg EPA and 20–32 mg/kg DHA for osteoarthritis, and considerably higher doses — around 225 mg/kg EPA and 90 mg/kg DHA — in the limited cognitive-decline literature. Those are body-weight-based numbers, which is exactly why “one scoop for all dogs” fish oil products are a red flag: a 12-pound Cavalier and an 80-pound Lab need very different amounts to hit a therapeutic range.
EPA in particular is doing most of the anti-inflammatory work in joint disease. Canine Arthritis Management notes that EPA is the primary omega-3 relevant to dogs with joint disease, with DHA carrying more weight for neurological and retinal tissue. That split matters when you’re comparing a label that just says “1,000mg fish oil” against one that breaks out EPA and DHA separately — only the second one lets you do the math against a published range. I had a ten-year-old Labrador, Otis, in for his senior wellness exam last spring — sixty-eight pounds, still food-motivated, but his owner had noticed him hesitating before jumping into the SUV, something he’d done without a thought a year earlier. We ran bloodwork, found nothing alarming, and I calculated an EPA/DHA dose against his weight rather than just handing over a bottle. The Counterargument I Take Seriously
The honest counterargument is that the NAD+ precursor data, while newer and thinner than the omega-3 literature, is a controlled trial with a real cognitive outcome measure — and cognition is exactly the domain where fish oil’s evidence is weakest, sitting at that higher, less-studied 225 mg/kg EPA range noted above. If a dog’s primary problem is disorientation, disrupted sleep-wake cycles, or slower recognition of familiar people, a targeted NAD+ precursor with a named dose and trial backing is a defensible choice, not a hopeful one.
Where I still land, though, is that most “vitality” complaints owners bring me aren’t pure cognition — they’re a blend of stiffness, lower energy, and a duller coat, and that combination has decades of osteoarthritis and cardiovascular data behind omega-3s that the NAD+ category simply hasn’t accumulated yet. Read the Label and the Bloodwork Before the Claim
This is the line I keep coming back to with every owner who sets two bags on my exam table: read the label and the bloodwork before the claim. A bag that says “cellular longevity” without naming milligrams of NR or NMN per chew isn’t giving you anything you can evaluate. A fish oil bottle that lists “1,000mg fish oil” instead of EPA and DHA separately is hiding the number that actually matters. Bloodwork matters just as much as the label. At the recheck, her coat had noticeably more shine and she was keeping pace on her usual loop around the block again. It’s one dog’s season, not a trial arm, but it’s the pattern I see often enough that I trust it: match the supplement category to the actual complaint, not to whichever bag has the more exciting front panel.
Neither category is a cure, and neither replaces a diagnosis. But if I had to choose one place to start for a dog whose “vitality” complaint is mostly physical — stiffness, energy, coat — it’s a properly dosed marine omega-3. If the complaint is genuinely cognitive — disorientation, disrupted sleep, slower recognition — the NAD+ precursor trial data gives you a real reason to consider that category instead. Read the label, read the bloodwork, and let the actual complaint pick the bag.
